Computational DNA Droplets Recognizing miRNA Sequence Inputs Based on Liquid–Liquid Phase Separation

Phase-separated biomolecular droplets are formed in cells to regulate various biological processes. This phenomenon can be applied constructing self-assembled dynamic molecular systems such as artificial and robots. Recently, programmable phase-separated called DNA have been reported a possible method construct systems. study reports computational droplet that recognize specific combination of tumor biomarker microRNAs (miRNAs) inputs output logic computing result by physical phase separation. A mixed consisting three nanostructures with orthogonal sticky-end sequences two linker DNAs cross-bridge the is proposed. By hybridization miRNAs linkers, cross-bridging ability lost, causing phase-separation into droplets, resulting executing miRNA pattern recognition described logical expression ((miRNA-1 ∧ miRNA-2) (miRNA-3 ¬miRNA-4)). experimentally demonstrates above chemically synthesized model experiment. In future, this will provide potential applications diagnosis therapy integration robots cells.
